Micromethod for rapid separation of lymphocytes from peripheral blood.

چکیده
The separation of lymphocytes from the other blood cells is based on the differences in their specific gravity. Centrifugation of whole blood against an isopaque-ficoll gradient results in the formation of three separate layers: the top one contains the slowly sedimenting cells (lymphocytes, platelets and some monocytes); the inter mediate one, the granulocytes; the bottom layer contains the erythrocytes, aggregated by the ficoll.

A new micromethod for evaluating lymphocyte responses to phytohemagglutinin: quantitative analysis of the function of thymus-dependent cells.
We have developed a new micromethod for testing phytohemagglutinin stimulation of human lymphocytes. 50 mul of peripheral blood is incubated at 37 degrees C directly, without cell separation, with 50 mul of tissue culture medium containing 50mug of phytohemagglutinin for 24 hr. Then, 0.5 muCi of tritiated thymidine in 50 mul of tissue culture medium is added and incubated for an additional 16 h...
متن کاملG2 chromosomal radiosensitivity and background frequency of sister chromatid exchanges of peripheral blood lymphocytes of breast cancer patients
Background: Chromosomal alterations play an important role in carcinogenesis. Enhanced chromosomal radiosensitivity is shown for many cancer predisposition conditions including breast cancer. In this study chromosomal radiosensitivity and the frequency of background sister chromnatid exchanges (SCE) in lymphocytes of normal individuals and breast cancer patients was compared. Materials...
متن کاملEffects of Gamma Irradiation on Proliferation and IL-5 Production of Peripheral Blood Lymphocytes
Gamma irradiation is routinely used for suppression of lymphocyte function in transfusion and transplantation procedures. In recent years, some investigators focused on the effects of ionizing radiation on special aspects of lymphocyte function and considered the possibility of its clinical application for treatment of some immunological disorders. In this study, we evaluated the effects of fiv...
متن کاملCombined Effects of 528 Hz Sound and X-ray in Peripheral Blood Lymphocytes
Introduction: Radiotherapy is still one of the main options for cancer treatment but it is in association with damage to normal cells as well as the tumor cells. To reduce the injury in normal cells we have evaluated the effect of 528 hertz sound after X irradiation in peripheral blood lymphocytes. Materials and Methods: in this study, peripheral blood was o...
متن کاملEffects of 35 Hz, 2 mT Magnetic Field on Peripheral Blood Lymphocytes of Human In Vitro and Rat In Vivo
In this research we have studied the effects of extremely low frequency magnetic field on the peripheral blood lymphocytes chromosomes of human in vitro and rat in vivo. By this means, we used 60 blood samples of 10 men as well as 40 rats. Of 60 human blood samples ,40 samples (in two group of 20) were selected as test groups and placed into the 2 mT and 35 Hz magnetic field. The blood samples,...
